The research team at Mediance Co., Ltd. has elucidated the core mechanism through which the transcription factor FOXO3a contributes to anti-aging effects and lifespan extension, and published its findings in the international peer-reviewed journal Biogerontology (IF 4.1, Top 25%). Using a genetically engineered mouse model designed to overexpress FOXO3a, the team demonstrated that FOXO3a regulates multiple biological processes, including enhanced DNA repair activity, reactive oxygen species removal, apoptosis regulation, stem cell activation, and autophagy induction. Additionally, the study confirmed that FOXO3a expression declines during aging, leading to decreased DNA repair capacity. Mechanistically, FOXO3a was shown to directly regulate DNA repair pathways at the transcriptional level. Importantly, in experiments exposing mice to high-dose gamma radiation (10 Gy), FOXO3a overexpression protected intestinal villus structure without adverse effects, reducing apoptosis and increasing both mean and maximum lifespan by approximately 30%. These findings indicate that FOXO3a plays a critical role in protecting cells against radiation-induced damage and improving survival outcomes. This study confirms that FOXO3a delays aging and promotes lifespan extension by enhancing DNA repair activity and regulating apoptosis. Based on this discovery, Mediance has initiated the development and screening of functional compounds that increase FOXO3a expression, with the goal of commercializing health functional supplements and pharmaceutical candidates targeting healthy longevity. Using a genetically engineered mouse model designed to overexpress FOXO3a, the team demonstrated that FOXO3a regulates multiple biological processes, including enhanced DNA repair activity, reactive oxygen species removal, apoptosis regulation, stem cell activation, and autophagy induction. Additionally, the study confirmed that FOXO3a expression declines during aging, leading to decreased DNA repair capacity. Mechanistically, FOXO3a was shown to directly regulate DNA repair pathways at the transcriptional level.
